Founded on December 12, 2001 by former player Luís Carlos Tóffoli, Cuiabá Esporte Clube began as an amateur academy before officially becoming a professional entity in 2003. The club faced temporary closure after winning its first state title in 2006 due to financial struggles, but Grupo Dresch revived the squad in 2009. This resurgence paved the way for a rapid climb through the Brazilian football hierarchy, culminating in a historic promotion to the Campeonato Brasileiro Série A in 2021. They became the first team from Mato Grosso to compete in Brazil's top flight since 1983.

The team plays its home matches at the Arena Pantanal, a 42,968-seat stadium constructed for the 2014 FIFA World Cup. In July 2024, the club opened the Manoel Dresch Training Center, described as one of the most modern facilities in the region. Currently competing in the Campeonato Brasileiro Série B following relegation from the top tier in 2023, the squad maintains a strong presence in the second division with recent victories against teams like América-MG and Vila Nova.

Cuiabá has established itself as a dominant force in local competition, holding the record for twelve Campeonato Mato-Grossense titles and claiming two Copa Verde championships. The squad values approximately €11.30 million consists entirely of domestic players, though midfielder Max Alves joined from the Colorado Rapids in early 2024.
